How IVD EIT™ works
The IVD EIT™ is the only nonanimal test method capable of discriminating all four GHS eye hazard categories.
The method is based on the principle that the depth of injury in the corneal tissue is the primary determinant of GHS eye hazard category. Superficial stromal injury corresponds to Category 2B or 2A; deeper injury extending into the deep stroma corresponds to Category 1. No stromal injury corresponds to Not Classified.
Tissues are processed into OCT-embedded tissue sections and stained for viability (Viability Stain Kit) or DNA fragmentation (TUNEL Kit). The depth of fluorescent staining in the corneal tissue directly predicts the GHS hazard category with greater than 90% accuracy.
IVD EIT is particularly valuable for pharmaceutical and excipient testing, where the full GHS classification spectrum is required and where the mechanistic relevance of a corneal tissue model is important.
Step-by-step protocol overview
Obtain Tissue
Fresh food-source tissue from certified supplier. Use within 24 hours of collection.
Corneal exposure
Overlay with tear, apply 100 µL of test material using dosing rings and exposure stabilizers from the Main Kit. Rinse with tear, incubate for 18 hours, fix tissue.
OCT processing
Embed the cornea in OCT compound and cryosection.
Stain sections
Apply Viability Stain Kit (IVDV3648) or TUNEL Kit (T3212).
Fluorescent imaging
Image sections using a fluorescent microscope.
Classify
Measure depth of injury and assign GHS category: NC, 2B, 2A, or 1.

Pair with Viability Stain Kit (IVDV3648) for live/dead staining, or TUNEL Kit (T3212) for DNA fragmentation detection.
Equipment check
A fluorescent microscope, OCT embedding equipment, and a cryostat or microtome are required.
